      目的:观察组蛋白去乙酰化酶抑制剂(HDACIs)MS-275 联合抗生素类化疗药物平阳霉素(PYM)对口腔鳞状癌细胞Tca-8113 的生长及凋亡的影响。方法:以体外培养的口腔鳞状细胞癌Tca-8113 细胞为研究对象,应用MTT 法检测不同浓度(0、1、2、4、8 μmol/L)MS-275、(0、0.05、0.1、0.2、0.4 μmol/L) 平阳霉素(PYM) 单独和联合用药对Tca-8113 细胞增殖活性的影响; Annexin-V-FITC/PI 双染流式细胞术定量检测细胞的凋亡情况。结果:不同浓度(1、2、4、8 μmol/L)MS-275 和(0.05、0.1、0.2、0.4 μmol/L)PYM 均可显著抑制Tca-8113 细胞的增殖,并促进其凋亡,且呈浓度依赖性(P<0.05)。4 μmol/L MS-275 和0.2 μmol/L PYM 联合应用时,其抑制Tca-8113 细胞增殖和促进其凋亡的作用均显著高于单独应用MS-275 或PYM(P<0.05)。结论:MS-275能有 效增强口腔鳞状细胞癌Tca-8113 细胞对平阳霉素(PYM)的敏感性,体外实验中呈现较好的抗肿瘤效应。

      Objective:To investigate the influence of combined application of histone deacetylase inhibitors MS-275 and antibiotics, chemotherapy drugs Pingyangmycin on the growth and apoptosis of oral squamous carcinoma cell Tca-8113.Methods:Oral squamous cell carcinoma Tca-8113 cells cultured in vitro were used for the study, MTT was performed to examine the proliferative activity of Tca-8113 cells after separated or combined application of (0, 1, 2, 4, 8μmol/L ) MS-275and (0, 0.05, 0.1, 0.2, 0.4 μmol/L) PYM; Annexin V-FITC/PI double dye, Flow cytometry was used to detect the effects of MS-275 plus PYMon the apoptosis of Tca-8113 cells.Results:Different concentrations(1, 2, 4, 8 μmol/L) of MS-275 or (0.05, 0.1, 0.2, 0.4 μmol/L) PYMcould both inhibit the growth and induce the apoptosis of Tca-8113 cells at a concentration dependent manner (P<0.05). Compared with MS-275 or PYM alone, 4 μmol/L MS-275 plus 0.2 μmol/L PYM could more significantly inhibit the proliferation of Tca-8113 cells and promote the apoptosis (P<0.05).Conclusion:MS-275 could effectively enhance the sensitivity of Tca-8113 cells to PYM, which showed good anti-tumour effect in vitro.
