      目的: 研究子痫前期患者胎盘组织中色素上皮衍生因子( pigment epithelium-derived factor, PEDF)的表达,探讨 PEDF 在子痫 前期发病中的作用。 方法: 选取 2012 年 3 月 至 2013 年 3 月 在我院产科住院剖宫产的 20 例 子痫前期孕妇作为 研究对象, 另 选取 同 期正常分娩的孕妇 20 例作为 对照组。 采用 Western blot、免疫荧光组织化学方法检测子痫前期患者和正常对照组妇女胎盘组织 中 PEDF 和血管内 皮生长因子 ( vascular endothelial growth factor, VEGF) 的表达, 并通过免疫荧光方法计数胎盘微血管密度 (MVD)。 结果: 同 正常对照组相比,子痫前期患者胎盘组织中 PEDF 表达升高, 而 VEGF 表达则 减少。 PEDF 与 VEGF 组织表达位 置大致相同 。子痫前期患者胎盘组织中 PEDF 表达与 24h 尿蛋白定量呈正相关, 与 VEGF 表达及 MVD 计数呈负 相关; VEGF 表 达与 MVD 计数呈正相关。 结论: PEDF 与 子痫前期疾病发生发展及病情轻重程度有关; PEDF 可能是通过影响胎盘血管的重铸, 而参与 子痫前期疾病的发生发展。

      Objective:To investigate the expression of pigment epithelium-derived factor (PEDF) placenta in of patients with preeclampsia, and to explore the role of PEDF in the pathogenesis of preeclampsia.Methods:20 cases of patients with preeclampsia who had elective cesarean deliveries in our hospital from March 2012 to March 201 3 were studied, and 30 women with normal term pregnancy were studied as the control group. The expression of PEDF, vascular endothelial growth factor (VEGF) and MVD in placentas were assayed using western blot and immuno fluorescence methods.Results:Compared with the normal control group, PEDF expression in placenta of preeclampsia patients increased, while the expression of VEGF decreased. PEDF and VEGF expression was roughly in the same position. The expression of PEDF in placenta of preeclampsia patients was positively correlated with 24h quantitative urinary protein, and negatively correlated with VEGF expression and MVD count. The expression of VEGF was positively correlated with MVD count.Conclusion:The expression of PEDF in placenta has correlation with the development and the severity of preeclampsia disease. PEDF may participate the occurrence and development of preeclampsia disease by influencing the blood vessels recasting in placenta.
