| 目的：检测p-p70S6K在结肠癌组织中的表达并探讨其临床意义。方法：选取40 例结肠癌组织蜡块以及40 例同一患者的正
常结肠组织蜡块进行免疫组化实验，其中又随机选取3组新鲜结肠癌组织和正常结肠组织，通过免疫印迹(Western blot)技术检测
p-p70S6K 在各组织中的表达情况。结果：在免疫组化实验中，癌组织阳性28 例，阴性12 例，阳性率为70%，正常结肠组织阳性14
例，阴性26 例，阳性率为35%，采用Pearson 卡方检验，得出x2=9.825，P=0.002＜0.05，说明癌组织与正常结肠组织中p-p70S6K的
表达差异有统计学意义；在免疫印迹实验中，以甘油醛-3- 磷酸脱氢酶(GAPDH) 为内参，重复试验三次，均显示目标蛋白
(p-p70S6K)分子量约70 KD，癌组织中p-p70S6K 表达较正常结肠组织明显增加,两组表达水平的比较采用t 检验,得出P=0.025＜
0.05，说明差异具有统计学意义。结论：p-p70S6K在结肠癌组织中异常表达，提示该分子在结肠癌的发生、发展过程中具有重要的
调控作用，进一步的研究可为结肠癌的靶向治疗提供分子生物学基础。 |

| Objective:To explore the role of p-p70S6K in the occurrence and development process of colon cancer.Methods:Immunohistochemistry was performed on the paraffin of normal colonic tissue and colon cancer tissue from the same patient of 40 cases,
and western blot was performed on 3 groups of fresh colon cancer tissue chosen at randomto detect the expression of p-p70S6K.Results:In immunohistochemistry, cancer tissue was positive in 28 cases, and negative in 12 cases, the positive rate was 70%; normal colonic
tissue was positive in 14 cases, and negative in 26 cases, the positive rate was 35%, Pearson chi-square test obtaining the x2=9.825, P=0.
002<0.05, showed that there were significantly different expression of p-p70S6K in cancer tissue and normal colonic tissue from the
identical patient; In Western blot test, GAPDH was chosen as the internal control, and the test repeated three times. Results showed that
the molecular weight of the target protein (p-p70S6K) was about 70 KD; and the expression of p-p70S6K in cancer was significantly
increased, compared with normal tissue of colon from the same patient, T test obtaining P=0.025<0.05, showed that there were
significantly different expression.Conclusion:The novel expression of p-p70S6K may be an important biologic factor in the occurrence
and development process of colon cancer, we may explore a potential target for treatment of colon cancer. |
