      目的:探讨高盐环境对小鼠脑缺血后IP-10(Interferon induced protein 10,IP-10)表达的影响。方法:将C57BL/6 雄性小鼠分 为高盐饮食(4%NaCl 鼠粮和1%NaCl 水)组和正常饮食组,建立永久性大脑中动脉结扎(permanent middle cerebral artery ligation, pMCAL)小鼠模型;激光共聚焦显微镜检测脑缺血组织IP-10 表达情况;RT-PCR(Reverse transcription PCR)检测脑缺血组织中 IP-10 表达情况;ELISA 方法检测内皮细胞和神经胶质细胞氧糖剥夺(oxygen-glucose deprivation,OGD)6h 后上清中IP-10 表达。 结果:免疫荧光观察发现高盐饮食组小鼠脑缺血组织中IP-10 阳性细胞表达增加;与正常饮食组相比,高盐饮食组缺血侧脑组织 中IP-10 表达显著增加,有统计学意义(P1d<0.05,P2d<0.001);与内皮细胞相比,神经胶质细胞OGD后,上清中IP-10 表达显著增 多(P<0.001)。结论:高盐环境促进脑缺血后IP-10 表达;脑缺血损伤过程中,脑内的IP-10 主要是由胶质细胞释放的。

      Objective:To study the effect of excess salt intake on the expression of IP-10 after cerebral ischemia.Methods:The experimental animal were divided into high salt diet group(4%NaCl rat food and 1%NaCl water)and normal diet group, then established the permanent middle cerebral artery ligation (pMCAL) model in mice; The expression of IP-10 in ischemic brain tissue was detected by laser scanning confocal microscope; The expression of IP-10 in cerebral ischemic tissues were detected by reverse transcription PCR; The expression of endothelial cells and glial cells in the supernatant of oxygen-glucose deprivation (OGD) 6 h were detected by ELISA.Results:The high salt group has more IP-10 positive cells vie immunofluorescence staining; Compared with the normal diet group, and the IP-10 expression level of high salt diet group mice was significantly increased by RT-PCR (P1d<0.05, P2d<0.001); Compared with the endothelial cells group, the IP-10 expression of glial cells was significantly increased in the supernatant after OGD (P<0.001).Conclusion:Excess salt intake promotes the expression of IP-10 after cerebral ischemia; endothelial cells and glial cells both express IP-10, and the expression of IP-10 of glial cells was higher than that of endothelial cells.
