      目的:探讨PECAM-1 在肝细胞肝癌(Hepatocellular carcinoma,HCC)组织中的表达及意义。方法:选择2013 年5 月-2015 年 6 月在我院接受治疗的HCC 患者100 例,收集肝癌患者HCC 组织及癌旁组织,另选取100 例正常肝脏组织作为对照组。应用免 疫组织化学法检测PECAM-1 在肝癌组织、癌旁组织以及正常肝脏组织中的阳性表达。利用小分子干扰RNA 技术(siRNA)构建 低表达的PECAM-1,并转染至肝癌细胞中抑制PECAM-1 的表达。应用Transwell小室法检测肝癌细胞的侵袭能力,CCK-8法检 测肝癌细胞的增殖能力。结果:PECAM-1 在肝癌组织、癌旁组织及正常肝脏组织中呈不同程度阳性表达(P<0.05);PECAM-1 在肝 癌组织及癌旁组织中的表达显著高于正常肝脏组织,差异具有统计学意义(P<0.05);PECAM-1 在肝癌组织中的表达显著高于癌 旁组织,差异具有统计学意义(P<0.05);转染siRNA PECAM-1 后,肝癌细胞中PECAM-1 mRNA的表达水平明显下降,PECAM-1 蛋白表达也明显降低,差异具有统计学意义(P<0.05);转染siRNA PECAM-1 后,肝癌细胞侵袭及增殖能力明显降低,差异具有统 计学意义(P<0.001)。结论:PECAM-1 在肝癌患者血清中高表达,PECAM-1 siRNA 能够抑制肝癌细胞的侵袭及增殖能力,提示 PECAM-1 可作为预测肝癌发生及发展的临床指标。

      Objective:To investigate the expression and significance of PECAM-1 in hepatocellular carcinoma (HCC).Methods:100 patients with hepatocellular carcinoma who had taken the resection surgery in our hospital from May 2013 to June 2015 were selected, and the HCC cancer tissues and the para-carcinoma tissues were collected. Another 100 normal liver tissues were collected as the control group. Then the expressions of PECAM-1 in the tumor tissues, the para-carcinoma tissues and the normal liver tissues were detected by RT-PCR. The small interfering RNA (siRNA) was used to inhibit the expression of PECAM-1 in hepatocellular carcinoma. The invasion of hepatocellular carcinoma cells was detected by Transwell, and the proliferation of hepatocellular carcinoma cells was detected by CCK-8.Results:PECAM-1 positively expressed in HCC tissues, adjacent tissues and normal liver tissues (P<0.05). The expressions of PECAM-1 in HCC tissues and adjacent tissues were significantly higher than that of the normal liver tissues (P<0.05). The expression of PECAM-1 in HCC tissues was significantly higher than that of the adjacent tissues (P<0.05). The level of PECAM-1 significantly decreased after transferred by PECAM-1 siRNA, and the invasion and proliferation of HCC cells significantly decreased, and the differences were statistically significant (P<0.05).Conclusion:PECAM-1 highly expressed in serum of patients with liver cancer, and siRNA PECAM-1 can inhibit the invasion and proliferation of HCC cells, that PECAM-1 can be used as a clinical indicator for predicting the occurrence and development of liver cancer.
