      目的:探讨尼可地尔预处理对大鼠缺血再灌注心肌mtDNA4834 bp 缺失突变的影响。方法:体重200 g-300 g,7-9 周龄健康 雄性Sprague-Dawley(SD)大鼠50 只,随机平均分为5 组,假手术(Sham)组、心肌缺血再灌注损伤(MIRI)组、缺血预处理(IPC)组、 尼可地尔(Nic)组、尼可地尔+5 羟基葵酸钠(Nic+5-HD)组。建立心肌缺血再灌注模型后取心肌组织,提取mtDNA,应用巢式PCR 法定性检测mtDNA4834 bp 片段缺失突变。结果:MIRI 组、IPC 组、Nic 组和Nic+5-HD 组突变频率分别为100%、20%、30%和 89%。Sham组未检测到mtDNA4834 bp 缺失突变。Nic 组mtDNA4834 bp 片段缺失明显低于MIRI组和Nic+5-HD组(P<0.05), 与IPC 组比较差异无统计学意义(P>0.05)。结论:尼可地尔预处理可降低大鼠MIRI 后mtDNA4834 bp 片段缺失突变,改善线 粒体能量衰竭。

      Objective:To investigate the effect of nicorandil preconditi oning on mitochondiral DNA 4834 bp deletion of myochondial ischemia-reperfusion injury in rats.Methods:Weighing 200 g-300 g, aged between 7-9 weeks fifty healthy male Sprague-Dawley(SD) rats were randomLy allocated into 5 groups(n=10 each): group Sham, group MIRI,group IPC, group Nic and group Nic+5-HD. The animals were sacrificed and myochondial tissues were collected, mitochondiral DNA was isolation. The mitochondrial DNA4834 bp deletion mutation was detected by nested-PCR (nested polymers chain reaction) technique.Results:The frequence of the mitochondrial DNA 4834 bp deletion mutation of MIRI group, IPC group, Nic group and Nic+5-HD group were 100%, 20%, 30%, 89%. The mtDNA4834 bp mutation frequency of Nic group was significantly lower than MIRI group and Nic+5-HD group (P<0.05), there were no significant difference between Nic group and IPC group (P >0.05).Conclusion:Nicorandil precondition decrease the mtDNA4834 bp deletion mutation of rats.
