      目的:探讨茶多酚对高糖诱导的人晶状体上皮细胞(human lens epithelial cells, HLECs)凋亡的影响。方法:建立高糖诱导的 HLECs 模型,用不同浓度的茶多酚干预,MTT比色法检测细胞存活率,电子显微镜观察细胞形态,透射电镜观察细胞内超微结构 变化,Western Blotting 检测凋亡相关蛋白的表达水平。结果:高浓度葡萄糖抑制了HLECs 活性,葡萄糖干预后细胞活性下降到 42.65%± 4.12%,与阴性对照组(96.07%± 4.02%)比较差异具有统计学意义(P<0.01)。用不同浓度茶多酚处理后,HLECs 活性分别 提高到55.33%± 4.15%,72.90%± 3.36%和76.00%± 3.79%,与氧化损伤组比较差异具有统计学意义(P<0.05);高浓度葡萄糖改变 了HLECs 形态及生长情况,而用茶多酚干预的高糖条件下的HLECs 则较好的保持了上皮细胞的形态;高浓度葡萄糖诱导 HLECs 凋亡反应的发生,引起凋亡相关蛋白表达水平改变,茶多酚抑制了Bax水平的升高,促进了Bcl-2 水平的下降。结论:茶多 酚可以抑制高糖诱导的HLECs 的凋亡,对糖尿病性白内障具有预防作用。

      Objective:To study the effect of tea polyphenols on high glucose induced-human lens epithelial cells (HLECs) apoptosis.Methods:High glucose-induced HLECs model was established, different concentrations of tea polyphenols were intervened, cells viability were assayed by MTT colorimetric, cells morphology were investigated by electron microscope, cell ultrastructural changes were detected by transmission electron microscopy, expression levels of apoptosis related proteins were tested by western blotting.Results:High glucose inhibited the activity of HLECs, after the intervention of the glucose cells activity decreased to 42.65%± 4.12%, the difference has statistical significance compared with negative control group (P<0.01). After treatment with different concentrations of polyphenols, HLECs activity were increased to 55.33%± 4.15%, 72.90%± 3.36% and 76.00%± 3.79%, the difference has statistical significance compared with oxidative damage group (P<0.01); high glucose changed HLECs morphology and growth station, while cell morphology of HLECs treated with tea polyphenols intervened in high glucose conditions were better maintained; high glucose induced HLECs apoptotic response, caused apoptosis-related protein expression levels change, tea polyphenols inhibited the levels of Bax elevated and Bcl-2 declined.Conclusion:Tea polyphenols can inhibit HLECs apoptosis induced by high glucose which have prevention effect in diabetic cataract.
