      目的:探讨Orexin-A 对肥胖大鼠摄食和自由活动的影响。方法:雄性SD 大鼠下丘脑外侧区(LH)置管,预先测量体重(BW), 脂体重(FM)和瘦体重(LM),据此对大鼠分组并分别提供高脂(HF)和低脂(LF)饮食饲喂,通过置管向大鼠LH 注射人工合成脑脊 液(aCSF)或orexin-A(OXA)。在饲喂前后监测OXA 对大鼠自发动态运动(SPA)以及摄食量,体重(BW),脂体重(FM)和瘦体重 (LM)的影响,以及每日LH 注射OXA是否能够抵抗高脂饮食引起的摄食诱导肥胖(DIO)。结果:高脂饮食饲喂之后,高摄食诱导 肥胖(DIO)大鼠和低DIO 大鼠间体重增加量(Δ BW)、摄食量和高脂饮食饲喂后的体重无差异(P>0.05),但与低脂饮食组大鼠相 比,上述各指标均显著增加(P < 0.05)。低DIO 大鼠的24 h SPA 和活跃期SPA 显著高于高DIO 大鼠(P < 0.05)。LH 注射OXA可 使严重DIO 降低而轻微DIO 增加,DIO 程度对LH 注射OXA 后的SPA 的影响呈非线性的。每日双侧LH 注射OXA 可抑制早期 DIO 但并不改变大鼠摄食量。LH 注射OXA 使大鼠SPA 增加,影响能量消耗,有助于DIO 抵抗。结论:Orexin-A 可通过增加大鼠 活动量,调控肥胖大鼠的摄食和DIO 抵抗作用。

      Objective:To investigate the effect of orexin-A on food take and spontaneous physical activity in obesity rats.Methods:Male Sprague-Dawley (SD) rats were prepared with unilateral cannulae targeting the lateral hypothalamus (LH) and orexin-A (OXA) or aCSF was injected as schedule. Rats were measured body weight (BW), fat mass (FM) and lean mass (LM), and classified into high-fat (HF) and low-fat (LF) diet groups according to above statistics. Then tested DIO effects on orexin signaling by measuring SPA after injection of OXA in the LH before and after 10 wk of HF feeding as well as whether daily OXA LH injections prevent DIO caused by HF feeding.Results:After dietary treatments, there were no differences between high-DIO and low-DIO rats In △BW, food intake, or BW after HF feeding, but both groups had significantly higher values compared with LF rats.(P < 0.05). Compare with high-DIO rats, there were significant higher of low-DIO rats in SPA over the 24 h and active periods(P < 0.05). We also found that development of severe DIO decreases whereas mild DIO increases after OXA injection in LH. The effects of DIO severity on SPA induced by OXA LH injections do not follow a linear trend. Daily injections of OXA in LH prevent DIO without altering food intake.Conclusion:OrexinA signaling contributes to the observed variations in SPA and DIO sensitivity.
