肿瘤外科确诊48 例直肠癌病理组织标本与20 例直肠癌患者癌旁正常组织标本为研究对象，应用免疫组化链霉菌抗生物素蛋白
-过氧化物酶连结法(S-P法)分析PTEN和Notch1 蛋白在直肠癌病理组织和正常组织中的表达情况。结果：PTEN 和Notch1主要
表达于直肠癌组织细胞质中，呈现棕黄色；Notch1 在直肠癌病理组织中表达率显著高于正常组织（P<0.05）；PTEN 在正常组织表
达率显著高于在直肠癌组织（P<0.05）；Notch1 在淋巴转移、中底分化及分期为C 期患者病理组织中表达率较高（P<0.05）；PTEN
在无淋巴转移、中底分化程度及A+B 期患者病理组织表达率较高（P<0.05）；相关性分析显示，Notch1和PTEN 在直肠癌病理组织
中表达呈现负相关关系（r=-0.534，P=0.000）。结论：Notch1 和PTEN 表达失调在直肠癌的发生、发展和淋巴结转移中有重要作用；
PTEN表达可能对直肠癌的发生、发展具有抑制作用，Notch1 表达可能对直肠癌的发生、发展具有促进作用。 |

| Objective:To explore the expression of PTEN and Notch1 in rectal cancer and their relationship with clinical
pathological parameters.Methods:48 cases of pathological tissues and 20 cases of carcinoma adjacent tissue were collected in oncology
department of our hospital from January 2014 to April 2015 as research objects; Immunohistochemical method of the anti biotin
peroxidase connection of streptomyces (S-P) was applied to analyse the expression of PTEN and Notch1 protein in colorectal cancer
tissues and normal tissues.Results:PTEN and Notch1 were mainly expressed in the cytoplasm of the cells of rectal cancer, showed a
brown yellow; The expression rate of Notch1 in colorectal cancer was significantly higher than that in normal tissues (P<0.05); The
expression rate of PTEN in normal tissues was significantly higher than that in colorectal cancer tissues (P<0.05); The higher expression
rate of Notch1 in lymph node metastasis, middle and bottomdifferentiation and C stage patients was higher (P<0.05); The expression rate
of PTEN was higher in the patients with no lymph node metastasis, middle and bottom differentiation and A+B stage (P<0.05);
Correlation analysis showed that the expression of Notch1 and PTEN in colorectal cancer was negatively correlated (r=-0.534, P=0.000).Conclusion:The dysregulation of PTEN and Notch1 expression may be associated with the occurrence and development of colorectal
cancer and lymph node metastasis. PTEN expression has probable inhibitory effect on the occurrence and development of colorectal
cancer and Notch1 expression can probably promote the occurrence and development of rectal cancer. |
