      摘要 目的:探讨食管鳞状细胞癌(Esophageal squamous cell carcinoma,ESCC)组织中线粒体单链DNA结合蛋白(single-strand DNA-binding protein 1,SSBP1)与线粒体转录因子A(mitochondrial transcription factor A,mtTFA)的表达及其临床病理意义。方法:采用免疫组化染色技术检测40例食管鳞状细胞癌组织与20例正常食管组织中SSBP1与mtTFA蛋白的表达,并分析其与食管鳞状细胞癌临床病理特征的相关性。结果:食管鳞状细胞癌组织中SSBP1和mtTFA的表达阳性率均明显高于正常食管组织( x2=9.909,P=0.002; x2=23.177,P=0.000)。SSBP1、mtTFA蛋白的表达与食管鳞状细胞癌的临床分期( x2=6.506,P=0.039; x2=7.535,P=0.023)、淋巴结转移( x2=7.305,P=0.007; x2=4.815,P=0.028)显著相关;mtTFA蛋白的表达与食管鳞状细胞癌的浸润深度有关( x2=8.541,P=0.014),SSBP1的表达与浸润深度无关( x2=5.695,P=0.058)。食管鳞状细胞癌组织中SSBP1与mtTFA的表达呈正相关(r=0.392,P=0.012)。结论:食管鳞状细胞癌组织中SSBP1和mtTFA蛋白均呈表达上调,二者可能协同作用参与食管鳞癌的发生、发展。

      ABSTRACT Objective: To investigate the expression and clinicopathological significances of mitochondrial DNA binding protein 1 (SSBP1) and mitochondrial transcription factor A (mtTFA) in the esophageal squamous cell carcinoma (ESCC) tissues. Methods: Immunohistochemical staining was used to detect the expression of SSBP1 and mt-TFA protein in 40 cases of esophageal squamous cell carcinoma and 20 normal esophageal tissue. Their correlation with the clinicopathological features of ESCC were analyzed. Results: The positive rates of SSBP1 and mtTFA in the ESCC were significantly higher than those in the normal esophageal tissues ( x2=9.909, P=0.002; x2=23.177, P=0.000). The expression of SSBP1, mtTFA protein were significantly related to the clinical stage of esophageal squamous cell carcinoma ( x2=6.506, P=0.039; x2=7.535, P=0.023), lymph node metastasis ( x2=7.305, P=0.007; x2=4.815, P=0.028). The expression of mtTFA protein was related to the depth of invasion of esophageal squamous cell carcinoma ( x2=8.541, P=0.014). The expression of SSBP1 showed no relationship with the depth of invasion ( x2=5.695, P=0.058). There was a positive correlation between SSBP1 and mtTFA expression in esophageal squamous cell carcinoma (r=0.392, P=0.012). Conclusion: SSBP1 and mtTFA proteins are up-regulated in the esophageal squamous cell carcinoma, and they may participate in the development and progression of esophageal squamous cell carcinoma.
