      目的:探讨环磷腺苷效应元件结合蛋白1(CREB1)及磷酸化环磷腺苷效应元件结合蛋白1(p-CREB1)在乳腺癌组织中的表达及其临床意义。方法:采用免疫蛋白印迹实验检测10例新鲜乳腺癌组织和邻近正常组织中的CREB1和p-CREB1的表达水平;采用免疫组织化学染色检测96例乳腺癌组织和60例邻近正常组织中CREB1和p-CREB1的表达,并分析其表达与临床病理特征和预后的关系。结果:免疫蛋白印迹实验结果显示CREB1和p-CREB1在乳腺癌组织中的表达水平显著高于邻近正常组织(P<0.05);免疫组织化学染色结果显示,CREB1和p-CREB1在癌组织中的高表达率显著高于邻近正常组织(P<0.05)。CREB1和p-CREB1的表达与乳腺癌患者腋窝淋巴结转移有关(P<0.05)。CREB1的表达与患者的无病生存率(DFS)和总生存率(OS)有关(P<0.05),而p-CREB1与患者的DFS和OS无关(P>0.05)。结论:CREB1及其磷酸化形式p-CREB1在乳腺癌组织中高表达,CREB1表达与不良预后相关,有望成为乳腺癌风险预测和治疗的新靶点。

      Objective:To investigate the expression of cAMP-response element binding protein1 (CREB1) and phospho-cAMP-response element binding protein1 (P-CREB1) in breast cancer tissues and their clinical significance. Methods: Western blot assay was used to detect the expression levels of CREB1 and p-CREB1 in 10 cases of fresh breast cancer tissues and adjacent normal tissues. Immunohistochemical staining was used to detect the expression of CREB1 and p-CREB1 in 96 breast cancer tissues and 60 adjacent normal tissues,the relationship between its expressionand clinicopathological parameters and prognosis were analyzed.Results: Western blot results showed that the expression levels of CREB1 and p-CREB1 in breast cancer tissues were significantly higher than those in adjacent normal tissues (P<0.05). Immunohistochemical staining results showed that the high expression rate of CREB1 and p-CREB1 in cancer tissues were significantly higher than those in adjacent normal tissues (P<0.05).The expressions of CREB1 and p-CREB1 were correlated with axillary lymph node metastasis of breast cancer patients (P<0.05). The expression of CREB1 was associated with disease-free survival rate (DFS) and overall survival rate (OS) of the patient (P<0.05), while p-CREB1 was not associated with DFS and OS of the patient (P>0.05).Conclusions: CREB1 and its phosphorylated form p-CREB1 are highly expressed in breast cancer tissues, and CREB1 expression is associated with poor prognosis, which is expected to be a new target for risk prediction and treatment of breast cancer.
