      目的 基于Toll样受体4/髓样分化蛋白88/核因子Kb(TLR4/Myd88/NF-κB)信号通路机制探讨microRNA-140-5p对于膝关节骨性关节炎(KOA)大鼠滑膜损伤中的保护作用研究。方法 采用前交叉韧带横断法(ACLT)建立KOA大鼠模型。大鼠注射microRNA阴性对照和microRNA-140-5p。通过实时定量聚合酶链反应(RT-qPCR)检测KOA滑膜组织和滑膜细胞中microRNA-140-5p的表达。RT-qPCR和蛋白免疫印迹试验检测TLR4/Myd88/NF-κB信号通路相关蛋白的表达。通过实验观察KOA对大鼠滑膜细胞增殖和凋亡的影响。检测KOA滑膜组织及滑膜细胞中炎症因子的表达水平。结果 KOA滑膜组织和滑膜细胞中microRNA-140-5p表达下调。上调microRNA-140-5p可抑制KOA大鼠滑膜细胞的炎症反应和细胞凋亡,促进滑膜细胞增殖。上调microRNA-140-5p可使KOA大鼠的TLR4/Myd88/NF-κB信号通路失活。结论 上调microRNA-140-5p可通过TLR4/Myd88/NF-κB信号通路失活化抑制KOA大鼠滑膜细胞炎症反应和凋亡,从而保护滑膜损伤。

      Objective:To investigate the protective effect of microRNA-140-5p on synovial injury in rats with knee osteoarthritis (KOA) based on the mechanism of Toll-like receptor 4/ myeloid differentiation protein 88/ nuclear factor kB (TLR4/MyD88/NF-κB) signaling pathway.Methods:A KOA rat model was established by anterior cruciate ligament transection (ACLT).Rats were injected with microRNA-negative control and microRNA-140-5p.MicroRNA-140-5p expression in synovial tissue and synovial cells of KOA was detected by real-time quantitative polymerase chain reaction (RT-qPCR).TLR4/MyD88/NF-κB signaling pathway related protein expression was detected by RT-qPCR and Western blot assay.The effects of KOA on the proliferation and apoptosis of rat synovial cells were observed.The expression levels of inflammatory cytokines in KOA synovial tissue and synovial cells were detected.Results:MicroRNA-140-5p expression was down-regulated in KOA synovial tissue and synovial cells.The up-regulation of microRNA-140-5p inhibited the inflammatory response and apoptosis of synovial cells in KOA rats,and promoted the proliferation of synovial cells.The up-regulation of microRNA-140-5p can inactivate the TLR4/MyD88/NF-κB signaling pathway in KOA rats.Conclusion:The up-regulation of microRNA-140-5p inhibited the inflammatory response and apoptosis of synovial cells in KOA rats through the inactivation of TLR4/MyD88/NF-κB signaling pathway, thereby protecting the synovial injury.
